- Abstract: The preparation, characterization and evaluation of γ -cyclodextrin/steviolbioside ( γ -CD/Sbio) inclusion complex was evaluated. Sbio is a typical steviol glycoside, which is used as a sucrose substitute and has health benefits. To overcome Sbio's poor aqueous solubility and low bioavailability, γ -CD/Sbio was prepared using a co-evaporation method. The possible bonding mode of Sbio with α -CD, β -CD and γ -CD was identified using AutoDock software. Host molecular screening tests showed that γ -CD was the optimal host molecule. The solubility of Sbio increased 31 fold, with a 1:1 γ -CD/Sbio inclusion complex. HPLC, FT-IR, TGA, XRD, SEM and NMR were used to characterize the γ -CD/Sbio. Based on an in vitro test, γ -CD/Sbio was more stable than Sbio with cecal contents. In addition, in vivo pharmacokinetics were done. The Cmax and AUC of γ -CD/Sbio were 2.4 times higher than for Sbio. Overall, the solubility and oral bioavailability of Sbio was significantly improved after inclusion. The relative oral bioavailability of Sbio was increased by about 2.4 times after inclusion.
