<abstract abstract-type="graphical" xml:lang="en"> <title> <x xml:space="preserve">Abstract</x> </title> <p> <bold>Shaping up for an A</bold>: Adenine is the only canonical nucleobase that does not offer a third hydrogen‐bonding functionality at its Watson–Crick face, making it difficult to bind with high affinity. A 6‐ethynyl‐2‐pyridone binds more tightly and with greater sequence fidelity than thymine.
