- Abstract : Cover : Cell‐sorting unilocular adipocytes isolated from white adipose tissue imposes a significant technological challenge, partly due to size and fragile nature of these cells. Isolating adipocytes could enable analysis of the heterogeneity in nutrient uptake and metabolism observed in white adipose tissue. In this issue, Robino et al. describe single‐cell sorting of rhesus macaque adipocytes labeled with a fluorescent fatty acid and live‐cell indicators, using large‐particle flow cytometry. The cover image shows omental white adipose tissue of the rhesus macaque, labelled with 4, 4‐Difluoro‐5, 7‐Dimethyl‐4‐Bora‐3a, 4a‐Diaza‐s‐Indacene‐3‐Dodecanoic Acid, a fluorescent‐conjugated free fatty acid (green); Calcein Red‐Orange AM, a live cell indicator (red); Hoechst 33342, a nuclear stain (blue); and Isolectin GS‐IB4, Alexa FluorTM 647 conjugate, a blood vessel marker (magenta). Live adipose tissue explants were labeled ex vivo with the indicated dyes in the presence of human insulin, and visualized by confocal microscopy. The adipose tissue exhibits cell‐autonomous heterogeneity in fatty acid uptake, resulting in a mosaic pattern of green fluorescence. Note that some adipocytes appear live (red), but exhibit lower levels of green fluorescence. Green speckles on the surface of the central lipid droplets represent precursor micro‐lipid droplets.
