- Abstract : Objective: Traumatic brain injury (TBI) induces severe brain damage which results in motor dysfunction and the therapy is poorly satisfactory. This study was designed to detect the role of eIF5A in rats with TBI, and investigate the underlying mechanism. Methods: TUNEL staining and neurological severity score (NSS) were used to detect the neuronal death and the motor function after TBI. Quantitative real‐time polymerase chain reaction and western blot were used to determine the expression of eIF5A in the cortex at 7 days after TBI. eIF5A shRNA (eIF5A‐SH) was used to downregulate its expression in the cultured cortex neurons. Lastly, the regulating relation between eIF5A and survivin was investigated. Results: NSS score increased after TBI, especially on day 7, and the cell apoptosis was also induced, which was companied by the downregulated expression of eIF5A, while eIF5A‐SH increased neuronal apoptosis. Moreover, the interaction experiment confirmed the eIF5A‐SH can influence the expression of survivin. Conclusion: eIF5A could maintain neuronal survival in the cortex after TBI, and expression of survivin could be regulated by eIF5A, which might be the underlying mechanism in TBI.
