- Abstract : Background: Cytokines are capable of manipulating the tumour microenvironment supporting tumour growth. Interleukin (IL)‐8 and monocyte chemoattractant protein (MCP)‐1, shown to be produced by various tumours, can negatively affect prognosis. The production of cytokines by canine mast cell tumours (MCT) has not been reported. Hypothesis/objectives: We hypothesise that MCT cell lines produce IL‐8 and/or MCP‐1 in addition to other cytokines, and that their production can be modulated by the Janus kinase (JAK) inhibitor oclacitinib. This pilot study aims to investigate the production of IL‐8, MCP‐1 and nine additional cytokines in three canine MCT cell lines, and determine the effects of oclacitinib on their production. Methods and materials: Reverse transcriptase‐PCR was used to detect the expression of IL‐8 and MCP‐1 mRNA in three MCT cell lines (CoMS, CM‐MC1 and VI‐MC1). The supernatant of the cell lines was evaluated for the presence of 11 cytokines [IL‐2, ‐6, ‐7, ‐8, ‐10, ‐15 and ‐18, and MCP‐1, granulocyte‐macrophage colony‐stimulating factor (GM‐CSF), interferon (IFN)γ and tumour necrosis factor (TNF)α] by enzyme‐linked immunosorbent assay (ELISA). The IC50 of oclacitinib was identified for each cell line. ELISA was performed again to compare changes in IL‐8 and MCP‐1 in treated cell lines versus untreated controls. Results: Interleukin‐8 and MCP‐1 were produced by all MCT cell lines tested. L'interleukine (IL)‐8 et la protéine chimiotactique des monocytes (MCP)‐1, qui sont produites par diverses tumeurs, peuvent affecter négativement le pronostic. La production de cytokines par les mastocytomes canins (TCM) n'a pas été rapportée. Hypothèse/Objectifs: Nous émettons l'hypothèse que les lignées cellulaires MCT produisent de l'IL‐8 et/ou du MCP‐1 en plus d'autres cytokines, et que leur production peut être modulée par l'inhibiteur de Janus kinase (JAK) oclacitinib. Cette étude pilote vise à étudier la production d'IL‐8, de MCP‐1 et de neuf cytokines supplémentaires dans trois lignées cellulaires MCT canines, et de déterminer les effets de l'oclacitinib sur leur production. Matériels et méthodes: La transcriptase inverse‐PCR a été utilisée pour détecter l'expression de l'ARNm de l'IL‐8 et de la MCP‐1 dans trois lignées cellulaires MCT (CoMS, CM‐MC1 et VI‐MC1). Le surnageant des lignées cellulaires a été évalué pour la présence de 11 cytokines [IL‐2, ‐6, ‐7, ‐8, ‐10, ‐15 et ‐18, et MCP‐1, facteur de stimulation des colonies de granulocytes‐macrophages ( GM‐CSF), l'interféron (IFN)γ et le facteur de nécrose tumorale (TNF)α] par dosage immuno‐enzymatique (ELISA). La CI50 de l'oclacitinib a été identifiée pour chaque lignée cellulaire. ELISA a été effectué à nouveau pour comparer les changements d'IL‐8 et de MCP‐1 dans les lignées cellulaires traitées par rapport aux témoins non traités. Résultats: L'interleukine‐8 et la MCP‐1 ont été produites par toutes les lignées cellulaires MCT testées. L'oclacitinib a significativement diminué la libération d'IL‐8 dans la lignée cellulaire CoMS et de MCP‐1 dans CoMS et VI‐MC1 à des concentrations cliniquement pertinentes. De plus, l'oclacitinib a significativement diminué la prolifération des trois lignées cellulaires. Conclusions: L'interleukine‐8 et la MCP‐1 sont produites par des lignées cellulaires MCT canines. La modulation de leur production est possible avec l'oclacitinib. Resumen: Introducción: algunas citoquinas son capaces de modificar el microambiente del tumor y favorecer su crecimiento. La interleuquina (IL) ‐8 y la proteína quimioatrayente de monocitos (MCP)‐1, que se ha demostrado que son producidas por diversos tumores, pueden afectar negativamente el pronóstico. No se ha publicado información acerca de la producción de citoquinas en mastocitomas (MCT). Hipótesis/Objetivos: se planteó la hipótesis de que las líneas celulares de MCT producen IL‐8 y/o MCP‐1 además de otras citoquinas, y que su producción puede ser modulada por el inhibidor de la quinasa Janus (JAK), oclacitinib. Este estudio piloto tiene como objetivo investigar la producción de IL‐8, MCP‐1 y nueve citoquinas adicionales en tres líneas de células MCT caninas y determinar los efectos de oclacitinib en su producción. Métodos y materiales: Se utilizó transcriptasa inversa‐PCR para detectar la expresión de RNAm de IL‐8 y MCP‐1 en tres líneas celulares MCT (CoMS, CM‐MC1 y VI‐MC1). El sobrenadante de las líneas celulares se evaluó para determinar la presencia de 11 citoquinas [IL‐2, ‐6, ‐7, ‐8, ‐10, ‐15 y ‐18, y MCP‐1, factor estimulante de colonias de granulocitos‐macrófagos (GM‐CSF), interferón (IFN) γ y factor de necrosis tumoral (TNF) α] mediante ensayo inmunoabsorbente ligado a enzimas (ELISA). Se identificó la IC50 de oclacitinib para cada línea celular. Se realizó de nuevo ELISA para comparar los cambios en IL‐8 y MCP‐1 en líneas celulares tratadas versus controles no tratados. Resultados: todas las líneas celulares MCT probadas produjeron interleuquina‐8 y MCP‐1. Oclacitinib disminuyó significativamente la liberación de IL‐8 en la línea celular CoMS y de MCP‐1 en CoMS y VI‐MC1 en concentraciones clínicamente relevantes. Además, oclacitinib redujo significativamente la proliferación de las tres líneas celulares. Conclusiones: la interleuquina‐8 y la MCP‐1 son producidas por líneas celulares caninas MCT. La modulación de su producción es posible con oclacitinib. Zusammenfassung: Hintergrund: Zytokine können die Mikroumgebung von Tumoren manipulieren und dadurch das Tumorwachstum unterstützen. Interleukin (IL)‐8 und das Monozyten Chemoattractant Protein (MCP)‐1, welches wie bereits beschrieben von verschiedenen Tumoren produziert wird, kann sich negativ auf die Prognose auswirken. Die Produktion von Zytokinen durch Mastzelltumoren (MCT) des Hundes wurde bisher nicht beschrieben. Hypothese/Ziele: Wir stellten die Hypothese auf, dass MCT Zelllinien ‐ zusätzlich zu anderen Zytokinen ‐ IL‐8 und/oder MCP‐1 produzieren und dass ihre Produktion durch den Janus Kinase (JAK) Inhibitor Oclacitinib moduliert werden kann. Diese Pilotstudie zielt darauf ab, die Produktion von IL‐8, MCP‐1 und neun weiteren Zytokinen in drei MCT Zelllinien zu untersuchen und die Auswirkungen von Oclacitinib auf ihre Produktion zu bestimmen. Methoden und Materialien: Die Reverse Transkriptase PCR wurde eingesetzt, um die Exprimierung von IL‐8 und MCP‐1 mRNA in drei MCT Zelllinien (CoMS, CM‐MC1 und VI‐MC1) zu bestimmen. Der Überstand der Zelllinien wurde auf das Vorkommen von 11 Zytokinen [IL‐2, ‐6, ‐7, ‐8, ‐10, ‐15 und ‐18 sowie MCP‐1, Granulozyten‐Makrophagen Kolonie‐stimulierender Faktor (GM‐CSF), Interferon (IFN)γ und Tumornekrosefaktor (TNF)α] mittels Enzyme‐linked Immunosorbent Assay (ELISA) untersucht. Die IC50 von Oclacitinib wurde für eine jede Zelllinie identifiziert. Ein ELISA wurde wiederum durchgeführt, um Veränderungen zwischen IL‐8 und MCP‐1 bei behandelten Zelllinien im Vergleich zu unbehandelten Kontrollen zu vergleichen. Ergebnisse: Interleukin‐8 und MCP‐1 wurden von allen getesteten MCT Zelllinien produziert. Oclacitinib verminderte die Freisetzung von IL‐8 in der CoMS Zelllinie und von MCP‐1 in CoMS und VI‐MC1 Zelllinien in klinisch relevanten Konzentrationen. Weiters verminderte Oclacitinib die Proliferation von allen drei Zelllinien signifikant. Schlussfolgerungen: Interleukin‐8 und MCP‐1 werden von caninen MCT Zelllinien produziert. A interleucina (IL)‐8 e a proteína quimioatrativa de monócitos (MCP)‐1, produzida por vários tumores, pode afetar negativamente o prognóstico. A produção de citocinas pelos mastocitomas caninos (MCT) ainda não foi relatada. Hipótese/Objetivos: Nossa hipótese foi a de que as linhagens celulares de MCT produzem IL‐8 e/ou MCP‐1 além de outras citocinas, e que a sua produção pode ser modulada pelo inibidor de Janus Kinase (JAK) oclacitinib. Este estudo piloto teve o objetivo de investigar a produção de IL‐8, MCP‐1 e nove citocinas adicionais em três linhagens celulares de MCT canino, e determinar os efeitos do oclacitinib na sua produção.
