Abstract

Abstract : The cover image is based on the Review "Cellular response to β‐amyloid neurotoxicity in Alzheimer's disease and implications in new therapeutics". β‐amyloid (Aβ) is a specific pathological hallmark of Alzheimer's disease (AD), and understanding its neurotoxic mechanism is crucial for Aβ‐targeted drug development. Aβ peptides are produced by sequential proteolytic cleavage of amyloid precursor protein (APP) by β‐secretase and γ‐secretase. The γ‐secretase creates C‐terminal variations and, together with N‐terminal truncations, gives diverse Aβ sequences and lengths. Some Aβ species are more amyloidogenic than others, such as Aβ1‐42 . As displayed in the cover background (Aβ1‐42 ), their misfolded conformation can easily gain cytotoxicity and cause protein aggregation. This review discusses some of the most recent advances in understanding Aβ‐centric AD mechanisms and makes some prospects for promising anti‐Aβ strategies.
