- Abstract: In this study, we have reported the molecular information of chemokine-19 (Chem19) from striped murrel Channa striatus ( Cs ). Cs CC-Chem19 cDNA sequence was 555 base pair (bp) in length which is 68 bp 5′ untranslated region (UTR), 339 bp translated region and 149 bp 3′ UTR. The translated region is encoded for a polypeptide of 112 amino acids. Cs CC-Chem19 peptide contains a signal sequence between 1 and 26 and an interleukin (IL) 8 like domain between 24 and 89. The multiple sequence alignment showed a 'DCCL' motif, an indispensable motif present in all CC chemokines which was conserved throughout the evolution. Phylogenetic tree showed that Cs CC-Chem19 formed a cluster with chemokine 19 from fishes. Secondary structure of Cs CC-Chem19 revealed that the peptide contains maximum amount of coils (61.6%) compared to α-helices (25.9%%) and β-sheet (12.5%). Further, 3D analysis indicated that the cysteine residues at 33, 34, 59 and 75 making the disulfide bridges as 33 = 59 and 34 = 75. Significantly (P < 0.05) highest Cs CC-Chem19 mRNA expression was observed in blood and it was up-regulated upon fungus and bacterial infection. Utilizing the coding region of Cs CC-Chem19, recombinant CsCC-Chem19 protein was produced. The recombinant Cs CC-Chem19 protein induced the cellular proliferation and respiratory burst activity of C. striatus peripheral blood leukocytes (PBL) in a concentration dependent manner. Highlights: Murrel 6C CC chemokine-19 characterized at molecular level. Cs CC-Chem19 contains an interleukin (IL) 8 like domain. Cs CC-Chem19 was highly expressed in blood upon microbial pathogen.
