- Abstract : Objectives: Oregovomab is an investigational murine monoclonal antibody directed against Cancer Antigen 125(CA-125), currently in clinical trials for treatment of patients with ovarian cancer expressing the tumor-associated cancer antigen CA-125. Outcomes are poor with non-platinum single agent chemotherapy in Poly-ADP Ribose Polymerase (PARP) inhibitor and platinum resistant setting. The aim of this study is to determine the efficacy of the indirect immunization with oregovomab in combination with non-platinum single agent chemotherapy in PARP inhibitor-resistant recurrent ovarian cancer patients not candidate for platinum retreatment. Methods: This is a prospective, multi-cohort, multicenter, phase II trial to evaluate the efficacy and safety of oregovomab in combination with non-platinum single agent chemotherapy in patients with recurrent epithelial ovarian cancer who have progressed with prior PARP inhibitor treatment and not candidate for platinum re-challenge. Patients were assigned to one of the followings: Cohort 1 (prior 1–3 lines of therapy), PLD (40 mg/m 2 q4w till PD) + Oregovomab 2 mg (C1, 2, 3, 5, 7 for 5 doses) or Cohort 2 (>3 prior lines of therapy), weekly paclitaxel (80 mg/m 2 D1, 8, 15 q4w till PD) + Oregovomab 2 mg (C1, 2, 3, 5, 7 for 5 doses). A total 56 subjected will be enrolled from 4 sites in South Korea. The primary endpoint is objective response rate. Accrual is expected to be started in July 2022.
