- Abstract : Phenotypic screening methods have placed numerous preclinical candidates into the antimalarial drug-discovery pipeline. As more chemically validated targets become available, efforts are shifting to target-based drug discovery. Here, we briefly review some of the most attractive targets that have been identified in recent years. Highlights: Many antimalarial drug targets discovered using reverse chemical genetic methods are druggable in other species. tRNA synthetases seem to be one of the most attractive and druggable classes of targets. Different druggable sites can be found for highly selective targets. Highly selective, target specific compounds may have resistance liabilities that may offset high potency and low toxicity.
