- Highlights: PF11 attenuates myocardial ischaemia injury and preserve heart function. PF11 regulates cardiac performance from inhibiting the expression of β1-AR. Molecular docking verified the binding sites between PF11 and β1-AR. Abstract: The anti-myocardial ischaemic effect of pseudoginsenoside F11 (PF11 ), a characteristic constituent of American ginseng, was evaluated in rats with coronary artery ligation. Both data and histopathological examination demonstrated that intragastric administration of PF11 at doses of 6 and 12 mg/kg could significantly improve heart function and decrease infarct size. Meanwhile, the levels of creatine kinase (CK), lactate dehydrogenase (LDH), aspartate transaminase (AST), malondialdehyde (MDA), and cardiac troponin T (cTnT) decreased, while the activity of superoxide dismutase (SOD) increased. Flow cytometry and western blotting were used to explore the underlying mechanism, and results showed that the expression of beta1-adrenoceptor (β1-AR) was decreased by PF11 . Furthermore, Glide XP software (Schrödinger) for molecular docking unravelled the probable binding sites between PF11 and β1-AR. This study provides substantial evidence for the effect of PF11 on myocardial ischaemia, and indicates that PF11 might be a potential dietary supplement for treatment of myocardial ischaemia injury.
