- Abstract: A goal in aging research is to identify robust molecular changes that track "normal aging" and departures from it. The "epigenetic clock" tracks chronological age across tissues and, independent of age, predict multiple health outcomes suggesting that it reflects the pace of molecular aging. The mechanism for the epigenetic clock is hard to discover, in part because methylation biology is not fully known. Alternatively, we used circulating proteins to construct a biomarker clock. Proteins directly affect phenotypes and provide information on biological pathways. Using SOMASCAN, we measured 1306 proteins in 240 healthy men and women between 22 and 93 years. Using a P≤3.83x10-5 significance threshold, 197 proteins were positively and 20 proteins were negatively associated with age. Using elastic net, we developed proteomic signature of age that use 75 proteins and is highly correlated with chronological age (r=0.94) in a validation sample. The generalizability of our findings needs independent replication.
