Abstract

Abstract : Background: Second-generation antipsychotics (SGAs) are associated with severe metabolic side effects which may be due to disrupted actions of central leptin (LEP). We investigated the ability of olanzapine (OLA) to impair hypothalamic LEP-mediated regulation of fuel preference and food intake. Methods: Male, Sprague-Dawley rats received an acute intracerebroventricular (ICV) infusion of LEP or vehicle along with an acute injection of OLA or vehicle at the start of 12-h light and dark cycles. Metabolic parameters were recorded using indirect calorimetry apparatus for 24-h. Glucose tolerance tests were subsequently performed. Results: OLA treatment rapidly reduced respiratory exchange ratio (RER) in light and dark cycles. Similarly, LEP and OLA co-administration reduced RER in light and most notably in the dark cycle. ICV-LEP significantly lowered cumulative food intake in the dark cycle regardless of OLA treatment. Acute OLA treatment resulted in glucose intolerance regardless of LEP treatment (p less than.05). Conclusion: Acute OLA and LEP exposure reduce RER and promote fatty acid oxidation. Importantly, while acute OLA does not appear to override the hypophagic effects of central LEP, OLA may impair the glucoregulatory effects of LEP.
