- Abstract : Insulin resistance is associated with many physiological conditions, including type 2 diabetes mellitus, obesity, pregnancy, and excess growth hormone (GH). The mechanism of insulin resistance, however, is poorly understood. Recent studies have identified phosphatidylinositol (PI) 3-kinase as a signaling molecule involved in this process. Specifically, GH has been shown to trigger insulin resistance and to enhance the expression of the p85α regulatory subunit of PI 3-kinase in skeletal muscle. This study explores the hypothesis that GH directly causes insulin resistance by increasing p85α protein level. 3T3-L1 pre-adipocytes were treated with recombinant rat GH (rrGH). The protein level of p85α in whole cell lysate was measured by Western blotting. The data demonstrated that rrGH directly increases the expression of p85α in a dose-dependent manner in the 3T3-L1 fibroblasts. Furthermore, rrGH does not seem to influence the protein level of the p110 catalytic subunit of PI 3-kinase.
