Abstract

Abstract : Background: Persian cats are predisposed to chronic and severe dermatophytosis. Alterations to the cutaneous microbiota are one potential contributor to this predisposition. Objectives: To characterise the cutaneous and environmental fungal microbiota of Persian cats with chronic, severe dermatophytosis, and to compare the fungal microbiota of cats with and without dermatophytosis. Animals: Thirty‐six client‐owned cats, including 26 Persian cats and 10 domestic long hair (DLH) cats. Methods and materials: Skin and home environment swabs were collected from Persian cats with severe, chronic dermatophytosis as well as groups of healthy control cats (Persian and DLH). Sequencing of the internal transcribed spacer 1 (ITS1) region was performed in addition to ITS1 quantitative PCR and fungal culture. Results: Next‐generation sequencing (NGS) targeting the fungal ITS region detected Microsporum sp. DNA from all Persian cats diagnosed with dermatophytosis and from environmental samples of their homes. A significant difference in community structure was identified between cases and controls, largely resulting from the Microsporum spp. DNA in samples from affected cats. Persian cats with dermatophytosis do not exhibit decreased fungal diversity. NGS failed to identify dermatophyte DNA on two culture‐positive asymptomatic Persian controls and identified Trichophyton rubrum DNA from a culture‐negative asymptomatic Persian control. Conclusions: Aside from M . canis, our results indicate that an underlying fungal dysbiosis is not likely to play a role in development of dermatophytosis in Persian cats. Other explanations for predisposition to this disease, such as a primary immunodeficiency, ineffective grooming or unique features of Persian cat hair should be investigated. Objectifs: Caractériser le microbiote cutané et environnemental fongique des persans atteints de dermatophytose chronique sévère et comparer le microbiote fongique des chats avec et sans dermatophytose. Sujets: Trente six chats de propriétaires, incluant 26 persans et 10 européens à poils longs (DLH). Matériels et méthodes: Les écouvillons cutanés et environnementaux ont été prélevés de chats persans atteints de dermatophytose chronique sévère ainsi que de chats sains contrôles (persans et DLH). Le séquençage de l'ITS1 (internal transcribed spacer 1) a été réalisé en plus d'une PCR quantitative ITS1 et d'une culture fongique. Résultats: Le séquençage de dernière génération (NGS) ciblant la région ITS fongique a détecté l'ADN de Microsporum sp. pour tous les persans diagnostiqués avec dermatophytose et pour l'environnement de leur habitat. Une différence significative de structure de communauté a été identifiée entre les cas et les contrôles, résultant largement de l'ADN de Microsporum sp. dans les échantillons des chats atteints. Les persans avec dermatophytose ne présentaient pas de diminution de la diversité fongique. NGS n'a pas permis d'identifier l'ADN de dermatophyte sur deux cultures positives de persans contrôles asymptomatiques et a identifié l'ADN de Trichophyton rubrum sur un chat contrôle asymptomatique avec culture négative. Conclusions: Hormis pour M . canis, nos résultats indiquent que la dysbiose fongique ne semble pas jouer un rôle dans le développement de la dermatophytose chez le chat persan. D'autres explications de prédisposition à cette maladie, telle qu'une immunodéficience primaire, un toilettage inefficace ou des critères uniques aux poils des persans devraient être étudiés. Resumen: Introducción: los gatos persas están predispuestos a la dermatofitosis crónica y grave. Las alteraciones de la microbiota cutánea son un contribuyente potencial a esta predisposición. Objetivos: Caracterizar la microbiota fúngica cutánea y ambiental de gatos persas con dermatofitosis crónica severa y comparar la microbiota fúngica de gatos con y sin dermatofitosis. Animales: Treinta y seis gatos de propietarios particulares, incluidos 26 gatos persas y 10 gatos domésticos de pelo largo (DLH). Métodos y materiales: Se recogieron hisopos de la piel y del entorno doméstico de gatos persas con dermatofitosis crónica grave, así como de grupos de gatos de control sanos (persas y DLH). Se realizó la secuenciación de la región del espaciador transcrito interno 1 (ITS1) además de una PCR cuantitativa de ITS1 y el cultivo de hongos. Resultados: la secuenciación de próxima generación (NGS) dirigida a la región fúngica ITS detectó DNA de Microsporum sp . en todos los gatos persas diagnosticados con dermatofitosis y de muestras ambientales de sus hogares. Se identificó una diferencia significativa en la estructura de la comunidad entre casos y controles, en gran parte como resultado del DNA de Microsporum sp . en muestras de gatos afectados. Los gatos persas con dermatofitosis no presentan una diversidad fúngica disminuida. NGS no pudo identificar el DNA de dermatofitos en dos controles persas asintomáticos con cultivo positivo e identificó el DNA de Trichophyton rubrum en un control persa asintomático con cultivo negativo. Conclusiones: Aparte de M. canis, nuestros resultados indican que no es probable que una disbiosis fúngica subyacente juegue un papel en el desarrollo de dermatofitosis en gatos persas. Zusammenfassung: Hintergrund: Perserkatzen sind prädisponiert dazu, an einer chronischen und schweren Dermatophytose zu erkranken. Veränderungen des Mikrobioms der Haut sind möglicherweise an dieser Prädisposition beteiligt. Ziele: Eine Beschreibung der mykotischen Mikrobiota der Haut und der Umgebung von Perserkatzen mit chronischer, schwerer Dermatophytose und ein Vergleich der mykotischen Mikrobiota von Katzen mit und ohne einer Dermatophytose. Tiere: Sechsunddreißig Katzen in Privatbesitz, wobei es sich um 26 Perserkatzen und 10 Kurzhaarkatzen (DLH) handelte. Methoden und Materialien: Es wurden Tupfer von der Haut und der Wohnungen von den Perserkatzen mit einer schweren, chronischen Dermatophytose sowie von den Gruppen der gesunden Kontrollkatzen (Perser und DLH) genommen. Es wurde eine Sequenzierung der internen transkribierten Spacer 1 (ITS1) Region sowie eine ITS1 quantitative PCR und eine Pilzkultur durchgeführt. Ergebnis: Eine Next‐Generation‐Sequencing (NGS) Technologie, die auf die ITS Region der Dermatophyten abzielte, wurde durchgeführt und wies bei allen mit einer Dermatophytose diagnostizierten Perserkatzen und bei den Umweltproben aus ihren Wohnungen Microsporum sp. DNA nach. Es wurde ein signifikanter Unterschied bei der Besiedelung zwischen den Fällen und den Kontrollen identifiziert, die sich weitgehend durch die Microsporum sp. DNA der Proben von betroffenen Katzen ergab. Perserkatzen mit einer Dermatophytose zeigten keine verminderte Diversität der Pilze. Die NGS konnte keine Dermatophyten DNA bei zwei Kultur‐positiven Perser Kontrollkatzen identifizieren und fand Trichophyton rubrum DNA bei einer Kultur‐negativen asymptomatischen Perser Kontrollkatze. Schlussfolgerungen: Neben M.canis weisen unsere Ergebnisse darauf hin, das eine zugrundeliegende Dysbiose der Pilze bei der Entstehung der Dermatophytosen der Perserkatzen eher keine Rolle spielt. Objetivos: Caracterizar a microbiota cutânea fúngica e ambiental de gatos persas com dermatofitose crônica grave e comparar a microbiota fúngica de gatos com e sem dermatofitose. Animais: Trinta e seis gatos de clientes, incluindo 26 gatos persas e 10 gatos domésticos de pêlo longo (DLH). Métodos e materiais: Swabs da pele e do ambiente doméstico foram coletados de gatos persas com dermatofitose crônica severa, bem como de grupos de gatos saudáveis controle (persas e DLH). O sequenciamento da região do espaçador transcrito interno 1 (ITS1) foi realizado além da PCR quantitativa de ITS1 e cultura de fungos. Resultados: O sequenciamento de próxima geração (NGS) visando a região ITS do fungo detectou DNA de Microsporum sp. em todos os gatos persas com diagnóstico de dermatofitose e nas amostras ambientais de suas casas. Uma diferença significativa na estrutura da comunidade foi identificada entre casos e controles, em grande parte resultante do DNA de Microsporum sp. em amostras de gatos afetados.
