- Graphical abstract: Highlights: The mitochondrial Na + /Ca 2+, NCLX emerged recently to be a pro-survival protein, preventing mitochondrial Ca 2+ overload and cell death. NCLX is the major entry pathway for Na + into the mitochondria. Na + mitochondrial influx, mediated by NCLX, modulate mitochondrial metabolic activity. Recent studies suggest a potential harmful route of NCLX activity by permitting mitochondrial Na + toxicity and impairing cell metabolic activity. Abstract: NCLX, the mitochondrial Na + /Ca 2+ transporter is a key player in Ca 2+ signaling. However, its role in Na + signaling is poorly understood. In this review we focus on Na + signaling by NCLX, and discuss recent physiological and pathophysiological roles attributed to the Na + influx into mitochondria.
