Abstract

Abstract: Background: Sufoof Jawahar Mohra (SJM) is a classical Unani formulation consisting of herbs and mineral origin ingredients. It is indicated as tonic for vital organs. Present study aims to evaluate anxiolytic and antidepressant activity and safety of SJM. Methods: SJM was orally administered at three doses i.e., 6, 12 and 24 mg/kg bw once daily for 28 consecutive days in rats. Porsolt's forced swim test (FST) was conducted to evaluate antidepressant potential and imipramine (10 mg/kg; p.o.) was used as positive control. Anxiolytic activity was evaluated using open field exploration test (OFT) and novelty induced suppression of feeding latency (FL) test. Diazepam (3 mg/kg, p.o.) was used as positive control. 90-days toxicity was assessed as per OECD guideline 408. Results: SJM reduced immobility time in FST at all three tested dose levels i.e., 6 mg/kg (73.83 ± 16.95 s), 12 mg/kg (48 ± 15.96 s; P < 0.05), and 24 mg/kg (28 ± 10.44 s; P < 0.01). In OFT, reduction in mean faecal dropping was found statistically significant in diazepam group (P < 0.001) as well as all three SJM groups (P < 0.01 for 6 and 12 mg/kg; P < 0.001 for 24 mg/kg dose). Rearing frequency and distance travelled in SJM treated groups was significantly higher (P < 0.05 vs. vehicle control) at 24 mg/kg dose (1082.00 ± 200.30 cm). Feeding latency was reduced with SJM at all tested dose levels, however, statistically significant reduction was observed only at 24 mg/kg bw (P < 0.05). No treatment related changes were observed in 90-days toxicity study in rats up to highest tested dose of 120 mg/kg of SJM. Conclusion: SJM showed promising antidepressant activity at 12 and 24 mg/kg in FST model. Anxiolytic activity was observed only at 24 mg/kg dose while two lower tested doses were not found to alleviate anxiety in rats. SJM may be considered safe up to the highest tested dose of 120 mg/kg bw.
