- Evaluation of: Medarova Z, Pham W, Farrar C, Petkova V, Moore A: In vivo imaging of siRNA delivery and silencing in tumors. Nat. Med. 13(3), 372––377 (2007) [1] . A recent paper on the fabrication and biological evaluation of a multifunctional nanoparticle for image-guided siRNA delivery is evaluated here. Superparamagnetic iron-oxide nanoparticle conjugates containing a cell-penetration peptide and a near-infrared fluorescence-imaging probe have been used to deliver siRNA to tumors. The conjugates demonstrated enhanced uptake and gene silencing in vitro and in vivo . The conjugates were monitored by magnetic resonance imaging (MRI) and near-infrared fluorescence imaging techniques and provided visual evidence of tumor uptake and therapeutic efficacy. I evaluate the results of this study and discuss the challenges in the development of these multifunctional drug-delivery nanoplatforms.
