Abstract

Abstract : The front cover picture shows an artificial backbone "foldamer" mimic of a small, disulfide‐rich protein. Replacing just a fraction of the natural amino acid residues in the prototype sequence with unnatural building blocks leads to a variant that undergoes clean oxidative folding to form a tertiary structure that is indistinguishable from the native domain. Backbone alteration imparts added protection against degradation by proteolytic enzymes, thus suggesting that the method described is a valuable approach to engineering the properties of bioactive disulfide‐rich scaffolds.
