- Abstract: STIM1, an ER-located Ca2+ sensor, activates Orai1 channels upon Ca2+-storedepletion. Prior to this, STIM1 undergoes a sequence of conformational changes, which cannot be controlled individually with high spatiotemporal resolution. Ma et al. [1 ] used the power of optogenetic engineering to transfer light-sensitivity to STIM1 and precisely characterize individual STIM1 activation steps.
