- Abstract: Unique properties of the biological network are their own ability to convert different types and quantity of biochemical information into dynamically different responses of the same molecules, which results in distinct cell fate decisions. Signaling pathways and the cell cycle machinery follow common principles regulating such decision-making by integrating all-or-none and graded molecular activities. They also share genes encoding and decoding quantitative information between the two systems. For instance, early-response genes decipher upstream signal dynamics and function as the network hub at the restriction point and regulate irreversible progression into the S phase of the cell cycle. This review provides an overview of signal-dependent transcription and cell cycle regulation and presents a mathematical model integrating the two networks to better understand the mechanisms underlying cellular decision-making. Highlights: Dynamic regulation of signaling activity, gene expression, and cell cycle entry are reviewed. Signal-responsive genes decipher upstream signaling dynamics. c-Fos and c-Myc, representative signal-responsive genes, regulate cell cycle entry. A mathematical model connecting these subnetworks elucidates cell cycle regulation controlled by cancer signaling.
