- Abstract : DOI: 10.1002/elps.201900100 The cover picture shows MS strategies for identification of protein SUMOylation by aid of the mutant SUMO‐1 tagging approach induced by CRISPR/Cas9 technique. The mutant SUMO‐1 gene is edited and introduced into cells using the CRISPR/Cas9 technique, which encodes R residue to replace the original T95 site of SUMO‐1 protein (SUMO‐1 T95R ) to produce a KGG tag fusing within the SUMO‐1‐modified protein. Then the substrate proteins conjugated with SUMO‐1 T95R are enriched by co‐immunoprecipitation (co‐IP) against anti‐SUMO‐1 antibody. Following trypsin digestion, the specific KGG‐tagging peptides from SUMOylated proteins are efficiently enriched by IP using anti‐K‐ε‐GG antibodies, which improves MS identification for the SUMOylated sites.
