- Abstract: Mitochondria, as the energy factories of cells, not only supply the energy but also are considered as novel and promising therapeutic targets for diagnosis, treatment, and prevention of various diseases, especially the cancer. Despite the expectation to direct the therapeutic drugs into the mitochondria, the actual mission is very difficult because of the complex mitochondrial special bilayer structure and highly negative potential nature of membrane. Over the time, various organic and inorganic nanocarriers were developed to improve the therapeutic activity of mitochondria acting drugs, which is a favorable platform to navigate across the physiological barriers and to achieve selective delivery of therapeutic and diagnostic agents to mitochondria. Although both inorganic and organic nanocarriers share the same size-dependent features, namely smaller and uniform size, high surface to volume ratio, and efficient cellular uptake, the inorganic nanocarriers usually exhibit novel physical properties, such as localized surface plasmon resonances, photoluminescence, or super-paramagnetism which can be exploited in many potential applications, including imaging and sensing. Moreover, some non–drug-loaded nanocarriers and inorganic/organic hybrid nanomaterials with a fatal effect on cells by influencing the mitochondrial functions will also be included. In general, the targeted formulations exhibited enhanced pharmacological effect and better therapeutic effect than their untargeted counterpart both in vitro and in vivo . At the end of this review, challenge and perspectives of mitochondria targeting inorganic nanocarriers would be discussed. Highlights: Advantages of nanocarriers with mitochondrial targeting property are discussed. Therapeutic barriers of mitochondrial targeting nanocarriers are discussed.
