- Abstract: Recently, it was discovered that the nanostructure of the wing of cicadas has bactericidal property. The reason is estimated that intercellular fluid is effused due to stretching the cell membrane on the nanostructural surface. Therefore, it is expected as a new antibacterial agent. However, effusion of the intracellular fluid has not been observed directly. In this study, we confirmed the effusion of intercellular fluid by using fluorescent proteins expressed in E. coli . The fluorescence intensity decreased, then it disappeared finally after the cell attached on the cicada wing. Missing of fluorescence was observed within 5 minutes from the cell attached on the wing of cicada. In addition, we analyzed time dependence of fluorescence intensity change for the single cell. From this result, time dependence of fluorescence intensity change can be separated into three processes. It was suggested that the process of adhesion to the nanostructure, the process of gradual emission of the fluorescent protein, and the process of massive outflow of the fluorescent protein
