- Abstract: A new coumarin-derived hydrazone (1 ) has been developed as a "turn-on" fluorescent probe for the detection of Cu 2+ in two different sensing mechanisms. In anhydrous acetonitrile, the strong fluorescence response of1 to Cu 2+ was mainly attributed to the chelation-controlled C=N isomerization. Besides, S-donor is indispensable in protecting from the Cu 2+ -induced fluorescence quenching. In aqueous acetonitrile, 1 as a chemodosimeter can highly selectively sense of Cu 2+, which was ascribed to the Cu 2+ -promoted cyclization reaction affording the strong fluorescent cyclization product (2 ). The proposed cyclization reaction was confirmed by the single-crystal structure of2 . Furthermore, 1 was utilized for imaging intracellular Cu 2+ with good performance. Graphical abstract: Highlights: A fluorescent chemodosimeter for Cu 2+ based on Cu 2+ -promoted cyclization reaction was developed. The fluorescent product of the Cu 2+ -promoted reaction was characterized by single-crystal X-ray analysis. The aza-michael addition based mechanism of the Cu 2+ -promoted cyclization reaction was proposed. The chemodosimeter was utilized for imaging intracellular Cu 2+ with good performance. The fluorescent probe can sense Cu 2+ in two different sensing mechanisms.
