- Abstract : Objective: A quantitative trait locus (QTL) for stroke proneness between the kallikrein (Klk) and Mt1pa markers on rat chromosome 1 was previously identified, and to gain functional insights, congenic strains were constructed by introgressing a selected chromosomal segment from the stroke-prone (SHRsp) onto the stroke-resistant (SHRsr) spontaneously hypertensive rat genome (BCIC rats) and vice versa (BCIIE rats) (Rubattu S et al. Physiol Genomics 2006, 27: 108–113). In the present study we investigated cerebral and mesenteric small resistance artery structure and function in 6 SHRsr, 6 SHRsp, 4 BCIC rats and 4 BCIIE rats. All rats were sacrificed at 12 week of age, after administration for 6 weeks of a high sodium/low potassium diet, when hypertension was fully developed. Figure. No caption available. Design and method: Mesenteric and cerebral (basilar) small resistance arteries were dissected and mounted on a wire myograph. A concentration-response to acetylcholine (from 10–9 to 10–5 Mol/L) was evaluated in vessels precontracted with norepinephrine 10–5 Mol/L). Morphological parameters were also evaluated, according to Mulvany and Halpern technique, in particular the media to lumen ratio was calculated. The congenic strains had intermediate values, but the differences vs. parental strains were not statistically significant. Acetylcholine–induced vasodilatation in mesenteric small resistance arteries was clearly less in SHRsp compared with SHRsr. In BCIIE rats the vasodilation response was slightly greater than SHRsp (ANOVA:NS), while in BCIC rats the vasodilation response was significantly worse than in SHRsr (ANOVA: p < 0.05). Conclusions: At 12 weeks of age, after development of structural alterations in cerebral and mesenteric small arteries, the introgression of a QTL from SHRsp to SHRsr and vice-versa influence endothelial function in mesenteric small arteries. In cerebral small arteries the introgression of a QTL from SHRsp to SHRsr (BCIC rats) tends to worsen structural alterations in respect with the parental strain. … (more)
