Abstract

Abstract : Background: Hypertension in the diabetic patient markedly increases the risk and accelerates the course of cardiac disease, peripheral vascular disease, stroke, nephropathy, and retinopathy. This study examine whether high sucrose and fat diet in diabetic type 2 sprague dawley rat model induce hypertension. Methods: Fourteen male Sprague Dawley Rat aged 2–3 months old were included in this study. They were divided into two groups control and experimental group. Control group received normal diet. The experimental group received high sucrose, high fat, and high sodium diet for ten weeks and intraperitoneal streptozococin injection (30 mg/BW) in the second week. The injection was repeated in the third week with the same dose. Fasting blood glucose level was measured every two weeks by colorimetric method. Plasma insulin was determined by ELISA method at the end of study. Blood pressure was determined by tail cuff blood pressure recorder. Results: Significant elevation of fasting blood glucose (103.14 ± 16.70 to235.29 ± 27.72 mg/dl; p < 0.001) and blood pressure (128.14 ± 5.78 to 155.42 ± 9.30 mmHg; p < 0.001) was observed in experimental group. However, their body weight was not significantly increased (287 ± 43.10 vs 334 ± 46.10; p > 0.62). On the other hand there were no significant changes on fasting blood glucose (94.43 ± 12.9 to 89 ± 13.32; p = 0.241) and blood pressure (125.14 ± 4.09 to 128.42 ± 5.15; p = 0.065) was observed in control group. Insulin level was significantly elevated in experimental group compared to that of control group (0.56 ± 0.09 vs 0.044 ± 0.06 mU/L; p = 0.018). HOMA IR also increased significantly in experimental group compared to that of control group (1.49 ± 0.46 vs 5.33 ± 1.21; p < 0.001). Conclusion: High sucrose and high fatdietfor 10 weeks could induce hypertension in diabetes type 2 Sprague Dawley Rat model that similar to those of human characteristics.
