Graphical abstract: Highlights: Four new C21 steroidal glycosides were isolated from Cynanchum taihangense . Cytotoxicity effects on HL-60, THP1, and Caco2 cell lines were evaluated. Two compounds exhibited selective cytotoxicity on Caco2 or THP1, respectively. The structure-activity relationship was analyzed. Abstract: Four new C21 steroidal glycosides, cynataihosides A (1 ), B (2 ), C (5 ) and D (6 ), together with two known compounds, cynanoside J (3 ) and glaucoside H (4 ), were isolated from the 95% ethanol extract of Cynanchum taihangense . The aglycone of cynataihoside A (1 ) was also a new compound, and that of cynataihoside B (2 ) was unusual. Their structures were elucidated on the basis of 1D- and 2D- NMR spectroscopic data, HR-ESI–MS analysis and qualitative chemical methods. The compounds were subjected to detect the cytotoxicity against three human tumor cell lines (HL-60, THP1 and Caco2). Compound2 selectively showed potent cytotoxic activity on Caco2 cells with IC50 value of 1.23 μM, being more effective than the positive control (5-fluorouracil, IC50 = 6.76 μM). Compound5 was more sensitive to THP1 cell line than HL-60 and Caco2 cell lines with IC50 values of 7.85 μM. Moreover, the structure-activity relationship of the C21 steroidal glycosides on the cell lines was analyzed.
