- ABSTRACT: Papillary thyroid carcinoma is a common health problem across the world. The goal of this study is to diagnose and detect papillary thyroid carcinoma in human thyroid tissue. Tissue samples were collected from Ataturk University, Department of Pathology. Diagnosis was achieved using histopathological tests as a reference protocol. Samples were deparaffinized and 20 µm of each sample was located on a coverslip. All tissues were treated with light from 100 to 4000 cm −1 and the spectrum was treated with appropriate software. Orthogonal partial least square analysis was used to distinguish tumor and healthy tissues. Mapping was performed by Raman spectroscopy including two-laser beam by filtering Rayleigh scattering. This method allows the diagnosis of papillary thyroid carcinoma in human tissue without sample preparation, extraction, or any other pretreatment. This easy and inexpensive method does not require chemicals for diagnosis. Chemometric-assisted Raman spectroscopy provides a new perspective on qualitative determination of this disease.
