- Abstract : The inside back cover picture shows the reconstruction of the ancestral sequence of haloalkane dehalogenases. Ancestral enzymes exhibited significantly enhanced thermodynamic stability compared to extant ones, as well as higher specific activities with a preference for short, multisubstituted halogenated substrates. Ancestral sequence reconstruction represents a powerful strategy for constructing highly stable and active catalysts that can further serve as robust templates for directed evolution.
