Abstract

Abstract: Dolichos kilimandscharicus is a perennial herb. In Zimbabwe the tuber powder is used against various insect pests. Saponins from this plant have molluscicidal and fungicidal properties. The aims of this study were to investigate the antibacterial activity of saponins from the roots of D. kilimandiscarius against S. aureus and P. aeruginosa, and to evaluate the anticancer effects of D. kilimandscharicus on human leukemia HL-60 cells. Growth inhibition assay, combination with ampicillin and drug transport assays in bacteria were conducted. The root tuber extract exhibited antibacterial activity on both bacteria, but the saponin fraction was more effective. The plant extract enhances the efflux of ciprofloxacin in bacteria. The effects of four extracts, (acetone, hexane, DCM and water) of D. kilimandscharicus roots on HL-60 cells were determined using the dye exclusion assay. The extracts had doseand time-dependent anti-proliferative effects on HL60 cells. The dichloromethane extract was shown to be the most potent of the four extracts. The anticancer effects of the extracts were shown to be irreversible, especially at high concentrations. The antiproliferative activity of the DCM extract was enhanced by camptothecin. The roots of D. kilimandscharicus are, thus, a potential source of lead compounds with antibacterial and anticancer activities.
