Abstract

Abstract : The cover picture shows a general labeling scheme used to create a dual‐fluorophore labeled variant of a large cellular protein machine, p97, for folding studies. Dual unnatural amino acid incorporation was used to install azide moieties on the protein, which were subsequently used for site‐specific dye labeling by using bioorthogonal Click‐chemistry in a background of a large number of native cysteines. The scheme potentiates detailed biophysical studies of this and other large cellular proteins using single‐molecule Förster resonance energy transfer (FRET).
