- <abstract xml:lang="en" abstract-type="author" id="ab005"> <title> <x xml:space="preserve">Abstract</x> </title> <sec> <p id="sp005">Inflammatory bowel disease is less common in countries where there is a greater risk of exposure to helminths. In our study, we examined the modulatory effect of the laminated layer extracted from the cyst wall of a helminth parasite, <italic>Echinococcus granulosus</italic>, on dextran sulfate sodium (DSS)-induced colitis in mice. Crude extract of <italic>E. granulosus</italic> laminated layer was injected intra-peritoneally daily, starting 3 days before colitis induction. The Disease Activity Index was monitored daily and histological scores were evaluated. In addition, the colonic expression of inducible nitric oxide synthase (iNOS) and nuclear factor-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgj2n6x7k97" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si5.gif" display="inline" overflow="scroll" id="d13e103" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi mathvariant="normal">κ</mml:mi></mml:mrow></mml:math></alternatives></inline-formula>B (NF-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgj2n6x7k97" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si5.gif" display="inline" overflow="scroll" id="d13e108" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi mathvariant="normal">κ</mml:mi></mml:mrow></mml:math></alternatives></inline-formula>B) was examined. Importantly, our results indicate that hydatid laminated layer pretreatment significantly improved the clinical symptoms and histological scores observed during DSS-induced colitis. Furthermore, treatment with hydatid laminated layer caused a significant decrease in NO (<sup>∗∗</sup><bold><italic>p</italic></bold> &lt; <bold>0.001</bold>), <bold>IFN</bold>-<bold>γ</bold> (<sup>∗</sup><italic>p</italic> &lt; 0.01) and TNF-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgj2n6x7r88" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si3.gif" display="inline" overflow="scroll" id="d13e140" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi mathvariant="normal">α</mml:mi></mml:mrow></mml:math></alternatives></inline-formula> production (<sup>∗</sup><italic>p</italic> &lt; 0.05) and an increase in IL-10 production. These results were associated with localized down-regulation of iNOS and NF-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgj2n6x7k97" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si5.gif" display="inline" overflow="scroll" id="d13e153" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi mathvariant="normal">κ</mml:mi></mml:mrow></mml:math></alternatives></inline-formula>B expression. Our results demonstrate the potent protective effect of hydatid laminated layer in DSS induced colitis through downregulation of IFN-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgj2n6x7mmq" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si1.gif" display="inline" overflow="scroll" id="d13e159" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi mathvariant="normal">γ</mml:mi></mml:mrow></mml:math></alternatives></inline-formula> and TNF-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgj2n6x7r88" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si3.gif" display="inline" overflow="scroll" id="d13e164" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi mathvariant="normal">α</mml:mi></mml:mrow></mml:math></alternatives></inline-formula> and up regulation IL-10 production. Furthermore, hydatid laminated layer played a beneficial role in maintaining the integrity of the intestinal mucosal barrier against DSS-induced injury suggesting the crucial role of helminth in immuno-protection against IBD.</p> </sec> </abstract> … (more)
