- <abstract xml:lang="en" abstract-type="author" id="ab005"> <title> <x xml:space="preserve">Abstract</x> </title> <sec> <p id="sp005">Bidirectional tumor–stroma interactions have tumor-promoting effects, and heterogeneity of carcinoma-associated fibroblasts (CAFs) has been showed, but its functional relevance for tumor development and invasion is still poorly understood. This study identified two CAF subtypes in oral squamous cell carcinoma (OSCC) based on transcriptome analysis. CAF-N were characterized by a secretome closer to NFs, included a significantly higher proportion of motile fibroblasts, and showed enhanced migration in response to TGF-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgjcwgwp8h" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si4.gif" overflow="scroll" id="d13e131" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi>β</mml:mi></mml:mrow></mml:math></alternatives></inline-formula>1. Migratory enhanced NFs (by TGF-<inline-formula><alternatives><inline-graphic xlink:href="ark:/27927/pgjcwgwnb2" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /><mml:math altimg="si5.gif" overflow="scroll" id="d13e136" xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mi>β</mml:mi></mml:mrow></mml:math></alternatives></inline-formula>1-pretreated) induced invasive properties in transformed oral keratinocytes, while inhibition of CAF-N migration impaired invasion of malignant oral keratinocytes, showing the essential role for fibroblast motility in oral carcinoma invasion supported by CAF-Ns.
