- <abstract abstract-type="main" id="icad12118-abs-0001"> <title>Abstract</title> <p> <list id="icad12118-list-0001" list-type="order"> <list-item> <p>Accurate identification of species of parasitoid Hymenoptera often requires the analysis of multiple genetic and non‐genetic traits.</p> </list-item> <list-item> <p>Here, we investigate the potential for nuclear polydnavirus (PDV) gene loci to provide species‐level discrimination in the parasitoid wasp genus <italic>Hyposoter</italic> (Hymenoptera: Ichneumonidae). A region of one PDV gene, <italic>Cys‐d9.2</italic>, was sequenced from nine species of wasps and an additional two PDV genes, <italic>Cys‐d9.1</italic> and <italic>Rep‐c18.2</italic>, were sequenced from multiple specimens of one species of wasp.</p> </list-item> <list-item> <p>A Bayesian phylogenetic analysis of the <italic>Cys‐d9.2</italic> sequences resulted in accurate identification of species and no intraspecific variation was observed in this gene, <italic>Cys‐d9.1</italic> or <italic>Rep‐c18.2</italic>. Further statistical analyses showed that <italic>Cys‐d9.2</italic> has a high prevalence of non‐synonymous nucleotide substitutions.</p> </list-item> <list-item> <p>Our results support the use of <italic>Cys‐d9.2</italic> as an additional genetic locus for species delimitation in <italic>Hyposoter</italic>, highlighting the value of PDV gene information to taxonomists studying the ichneumonid subfamily, Campopleginae.</p> </list-item> </list> </p> </abstract>
