- <abstract xml:lang="en" abstract-type="author" id="abs0010"> <title id="sectitle0010">Abstract</title> <sec> <p id="abspara0010">Toll-like receptors (TLRs) play an important role in regulation of anti-microbial peptides (AMPs) expression. A novel vertebrates TLR counterpart named <italic>PcToll</italic>, was firstly identified from the freshwater crayfish, <italic>Procambarus clarkii</italic>. Phylogenetic analysis showed that PcToll together with <italic>Drosophila melanogaster</italic> and <italic>Anopheles gambiae</italic> Toll9 were clustered with human Tolls. <italic>PcToll</italic> was mainly expressed in hepatopancreas and gills and it also could be detected in hemocytes, heart, stomach and intestine. <italic>PcToll</italic> was upregulated in hemocytes and gills post 24 h <italic>Vibrio anguillarum</italic> challenge. In hepatopancreas and intestine, the highest expression level of <italic>PcToll</italic> could be observed at 12 h <italic>V. anguillarum</italic> challenge. In hemocytes, <italic>PcToll</italic> went up post 24 h <italic>Staphylococcus aureus</italic> challenge and in gills, the expression level of <italic>PcToll</italic> showed no obvious change from 2 to 24 h <italic>S. aureus</italic> challenge. In hepatopancreas post 12 h <italic>S. aureus</italic> challenge, <italic>PcToll</italic> was upregulated and it showed obvious upregulation post 12 h <italic>S. aureus</italic> challenge in intestine. Overexpression of <italic>PcToll</italic> in <italic>Drosophila</italic> S2 cells could induce <italic>Drosophila Attacin</italic> (<italic>Atta</italic>), <italic>Metchnikowin</italic> (<italic>Mtk</italic>), <italic>Drosomycin</italic> (<italic>Drs</italic>) and shrimp <italic>Penaeidin</italic> (<italic>PEN4</italic>) expression.
