Abstract

<abstract> <title> <x xml:space="preserve">Abstract</x> </title> <p>Polymorphisms in <italic>ERCC1</italic>, <italic>XPD</italic>, and <italic>XRCC1</italic> were examined for (a) association with the clinical outcome of 107 non-small cell lung cancer patients receiving front-line platinum-based chemotherapy, and (b) correlation with the <italic>ERCC1</italic> mRNA levels of 176 chemo-naive primary tumors. The <italic>ERCC1-C8092</italic> allele and the number of <italic>ERCC1</italic> polymorphic variants (<italic>C8092A</italic> and <italic>Asn118Asn</italic>) were associated with progression-free survival. In non-squamous histology, tumoral <italic>ERCC1</italic> mRNA levels were lower in patients homozygous for <italic>ERCC1-C8092</italic> as compared with the patients carrying the A allele (<italic>p</italic> = .024). These findings merit investigation in larger cohorts of patients treated with uniform regimens.</p> </abstract>
