- <abstract abstract-type="main"> <title> <x xml:space="preserve">Abstract</x> </title> <p> <underline> <bold>Background:</bold> </underline> Although Ncor1 and Ncor2, the co‐repressors that can actively repress gene transcription through binding nuclear receptors in the absence of ligands, are crucial to vertebrate embryogenesis, their roles in its primitive myelopoiesis remain unknown. We investigated the function of <italic>ncor1</italic> or <italic>ncor2</italic> in zebrafish embryos by antisense morpholino knocking down technologies. <underline><bold>Results:</bold></underline> Development of both <italic>mfap4</italic><sup>+</sup> macrophages and <italic>mpx</italic><sup>+</sup> neutrophils was abolished in <italic>ncor2</italic> morphants, whereas development of <italic>mpx</italic><sup>+</sup> neutrophils was depleted in <italic>ncor1</italic> morphants. <italic>ncor2</italic> was essential to the development of <italic>spi1b<sup>+</sup></italic> myeloid precursors but not anterior hemangioblasts whereas <italic>ncor1</italic> was dispensable to the specification of <italic>spi1b<sup>+</sup></italic> myeloid precursors and anterior hemangioblasts. Overexpressing <italic>spi1b</italic> could partially rescue expressions of <italic>mfap4</italic> and <italic>mpx</italic> in <italic>ncor2</italic> morphants.
