- <abstract abstract-type="main" xml:lang="en"> <title>Abstract</title> <p>Bis(phenylethynyl)pyridylcarboxamides with amide side chains at the <italic>para</italic> position of the NH<sub>2</sub> group possess strong solvatochromic properties compared with the <italic>meta</italic> analogues. Fluorescence binding titrations show that these probes exhibit remarkable fluorescence turn‐on responses upon interacting with the human telomeric G‐quadruplex (<italic>h‐TELO</italic>). Förster resonance energy transfer melting analysis shows the high selectivity of these probes for <italic>h‐TELO</italic> over duplex DNA. Isothermal titration calorimetry, as well as UV/Vis and fluorescence spectroscopy studies, show that the <italic>meta</italic> analogue has a twofold binding affinity for <italic>h‐TELO</italic> over the <italic>para</italic> analogue. The noncovalent interaction of these small‐molecule probes with <italic>h‐TELO</italic> has been used to regulate the assembly of novel supramolecular nanoarchitectures.</p> </abstract>
