- <abstract abstract-type="main" id="age12188-abs-0001"> <title>Summary</title> <p>The aim of the current work was to analyze, in the Sarda breed goat, genetic polymorphism within the casein genes and to assess their influence on milk traits. Genetic variants at the <italic>CSN1S1</italic>, <italic> CSN2</italic>, <italic> CSN1S2</italic> and <italic>CSN3</italic> gene loci were investigated using PCR‐based methods, cloning and sequencing. Strong alleles prevailed at the <italic>CSN1S1</italic> gene locus and defective alleles also were revealed. Null alleles were evidenced at each calcium‐sensitive gene locus. At the <italic>CSN3</italic> gene locus, we observed a prevalence of the <italic>CSN3 A</italic> and <italic>B</italic> alleles; the occurrence of rare alleles such as <italic>CSN3 B'', C, C', D, E</italic> and <italic>M</italic>; and the <italic>CSN3 S</italic> allele (GenBank <ext-link ext-link-type="gen" xlink:href="http://www.ncbi.nlm.nih.gov/nuccore/KF644565?report=GenBank" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink">KF644565</ext-link>) described here for the first time in <italic>Capra hircus</italic>. Statistical analysis showed that all genes, except <italic>CSN3</italic>, significantly influenced milk traits. A relevant finding was that <italic>CSN2</italic> and <italic>CSN1S2</italic> genotypes affected milk protein content and yield. The polymorphism of the <italic>CSN2</italic> gene affected milk protein percentage with the highest values recorded in the <italic>CSN2 AA</italic> goats (4.35, at <italic>P </italic>&lt;<italic> </italic>0.001). The <italic>CSN1S2 AC</italic> goats provided the highest fat (51.02 g/day) and protein (41.42 g/day) (<italic>P </italic>&lt;<italic> </italic>0.01) production.
