- <abstract abstract-type="graphical" xml:lang="en"> <title> <x xml:space="preserve">Abstract</x> </title> <p> <bold>Stereoselective traffic engineering</bold> The cover picture shows how engineered <italic>S</italic>‐selective <italic>Ec</italic>MenD variants catalyze the carboligation of α‐ketoglutarate and benzaldehyde derivatives with excellent enantioselectivities of up to 99 % <italic>ee S</italic>, in contrast to the wild‐type enzyme, which produces <italic>R</italic>‐enantiomers. In their Full Paper on <ext-link ext-link-type="doi" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink">p. 3587 ff.</ext-link>, M. Pohl et al. describe how they engineer <italic>S</italic>‐selective <italic>Ec</italic>MenD variants by optimizing steric properties and stabilization of the acceptor substrate in the <italic>S</italic>‐pocket. This method, inspired by the recently developed <italic>S</italic>‐pocket concept, involves opening the <italic>S</italic>‐pocket with simultaneous destabilization of the <italic>R</italic>‐pathway to enhance the <italic>S</italic>‐selectivity of thiamine diphosphate‐dependent enzymes.<boxed-text content-type="graphic" position="anchor" orientation="portrait"><graphic position="anchor" mimetype="image" xlink:href="ark:/27927/pgg4t7hjcpj" orientation="portrait" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /></boxed-text></p> </abstract>
