- <abstract abstract-type="main" id="fml12317-abs-0001"> <title>Abstract</title> <p>Both <italic>Streptococcus mutans</italic> and <italic>Streptococcus sanguinis</italic> are normal bacterial inhabitants of dental plaque. <italic>Streptococcus mutans</italic> is the major agent causing dental caries. It has been well documented that nicotine affects the growth of <italic>S. mutans</italic>. This study investigated the effect of nicotine on mono‐ and dual‐species growth of <italic>S. mutans</italic> and <italic>S. sanguinis</italic>. The results indicate that nicotine has no significant effect on <italic>S. sanguinis</italic> grown in either mono‐ or dual‐species biofilms. However, nicotine significantly increased (<italic>P</italic> &lt; 0.05) the growth of <italic>S. mutans</italic> in dual‐species biofilm formation. In addition, the CFU level of <italic>S. sanguinis</italic> was higher than <italic>S. mutans</italic> without nicotine in the culture. With the addition of nicotine, the level of <italic>S. mutans</italic> biofilm was significantly enhanced as the nicotine concentration increased over the level of <italic>S. sanguinis</italic> in dual‐species biofilm, and we also got the same result from the fluorescence <italic>in situ</italic> hybridization detecting the two bacteria grown in biofilm formation.
