- <abstract abstract-type="graphical" xml:lang="en"> <title> <x xml:space="preserve">Abstract</x> </title> <p> <bold>To gain insight</bold> into the catalytic mechanism of non‐ribosomal amino acid transferases, peptidyl‐RNA conjugates were synthesized for co‐crystallization with FemX<sub>Wv</sub> of <italic>Weissella viridescens</italic>, which transfers <sc>L</sc>‐Ala from Ala‐tRNA<sup>Ala</sup> to the peptidoglycan precursor UDP‐MurNAc‐pentapeptide. The structure of the resulting complex and mutational studies revealed the mechanism by which FemX<sub>Wv</sub> binds its substrates for substrate‐assisted catalysis and stabilization of the tetrahedral intermediate.<boxed-text content-type="graphic" position="anchor" orientation="portrait"><graphic position="anchor" mimetype="image" xlink:href="ark:/27927/pgg26mfvgb2" orientation="portrait" xlink:type="simple" xmlns:xlink="http://www.w3.org/1999/xlink" /></boxed-text></p> </abstract>
