Abstract

<abstract abstract-type="graphical" xml:lang="en" id="humu22234-abs-0001"> <title> <x xml:space="preserve">Abstract</x> </title> <p>We developed cellular models recapitulating the occurrence of multiple cancer mutations to investigate their role in tumor progression. Knock in of <italic>EGFR</italic>, <italic>KRAS</italic>, <italic>BRAF</italic> or <italic>PIK3CA</italic> oncogenic variants was combined with knock‐down of <italic>PTEN</italic> or <italic>RB1</italic> in human non‐transformed epithelial cells. Different combinations of alterations resulted in distinct biochemical properties and anchorage‐independent growth abilities. Nevertheless, those genetic events were insufficient to fully transform target cells.
