<abstract abstract-type="graphical" xml:lang="en"> <title> <x xml:space="preserve">Abstract</x> </title> <p> <bold>The back cover picture depicts</bold> the beauty and significance of water molecules, key structural components in the architecture of living systems, for the rational design of small molecules with tailored biological activity. A potent and selective inhibitor of the therapeutically relevant target matrix metalloproteinase 13 was realized, without any screening activities, by a concise structure‐based approach targeting multiple water molecules as binding partners.
