- <abstract abstract-type="main" id="tpj12389-abs-0001"> <title>Summary</title> <p>Recessive strain‐specific resistance to a number of plant viruses in the <italic>Potyvirus</italic> genus has been found to be based on mutations in the eukaryotic translation initiation factor 4E (<italic>eIF4E</italic>) and its isoform, <italic>eIF(iso)4E</italic>. We identified three copies of <italic>eIF(iso)4E</italic> in a number of <italic>Brassica rapa</italic> lines. Here we report broad‐spectrum resistance to the potyvirus <italic>Turnip mosaic virus</italic> (TuMV) due to a natural mechanism based on the mis‐splicing of the <italic>eIF(iso)4E</italic> allele in some TuMV‐resistant <italic>B. rapa</italic> var. <italic>pekinensis</italic> lines. Of the splice variants, the most common results in a stop codon in intron 1 and a much truncated, non‐functional protein. The existence of multiple copies has enabled redundancy in the host plant's translational machinery, resulting in diversification and emergence of the resistance. Deployment of the resistance is complicated by the presence of multiple copies of the gene. Our data suggest that in the <italic>B. rapa</italic> subspecies <italic>trilocularis</italic>, TuMV appears to be able to use copies of <italic>eIF(iso)4E</italic> at two loci.
