- <abstract> <title>Abstract</title> <p> <italic>Context</italic>: A methanol extract of <italic>Cyperus rotundus</italic> L. (Cyperaceae) rhizomes showed inhibitory activity against α-glucosidase and α-amylase, two enzymes involve in carbohydrate digestion.</p> <p> <italic>Objective</italic>: Identification of compounds from <italic>C. rotundus</italic> rhizomes responsible for the inhibition of α-glucosidase and α-amylase.</p> <p> <italic>Materials and methods</italic>: Compounds were identified by a phytochemical investigation using combined chromatographic and spectroscopic methods. α-glucosidase and α-amylase inhibitory activities were evaluated by <italic>in vitro</italic> enzyme inhibition assays.</p> <p> <italic>Results</italic>: A new (2<italic>RS</italic>, 3<italic>SR</italic>)-3, 4′, 5, 6, 7, 8-hexahydroxyflavane (<bold>1</bold>), together with three known stilbene dimers cassigarol E (<bold>2</bold>), scirpusin A (<bold>3</bold>) and B (<bold>4</bold>) were isolated. Compound <bold>2</bold> inhibited both α-glucosidase and α-amylase activities while the flavane <bold>1 </bold>only showed effect on α-amylase, and compounds <bold>3</bold> and <bold>4</bold> were active on α-glucosidase. All four compounds showed significant 2, 2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity.</p> <p> <italic>Discussion</italic>: The inhibitory activities against α-amylase and α-glucosidase of the <italic>C. rotundus</italic> rhizomes were reported for the first time.
