- <abstract abstract-type="main" xml:lang="en" id="efp12017-abs-0001"> <title>Summary</title> <p>Cytoskeletal dynamics play a crucial role in pathogen recognition and cell defence during the initial interactions between an invader and plant host. The aim of the work reported here was to characterize how <italic>Heterobasidion annosum</italic> s.s., <italic>Heterobasidion parviporum</italic>, and <italic>Heterobasidion abietinum</italic> affect the microtubules and microfilaments of <italic>Pinus sylvestris</italic> root cells 12‐, 24‐, 48‐, and 96‐h post‐inoculation. Inoculation of <italic>P. sylvestris</italic> with <italic>H. parviporum</italic> or <italic>H. abietinum</italic>, which have a lower specificity for <italic>P. sylvestris</italic> than <italic>H. annosum</italic> s.s, resulted in greater reorganization of host microtubules during the early stages of interaction than inoculation with the more specific <italic>H. annosum</italic> s.s. In some infected cells, spots of actin aggregates were observed. Disruption of cytoskeletal components by the application of specific cytoskeletal inhibitors facilitated the entry of the <italic>H. parviporum</italic> and <italic>H. abietinum</italic> into roots. These results suggest that the <italic>P. sylvestris</italic> cytoskeleton plays a role in the host response in the initial stages of the host–pathogen interaction.</p> </abstract>
