- <abstract abstract-type="main" xml:lang="en"> <title>Abstract</title> <sec id="mim514-sec-0001" sec-type="section"> <p>In Japan, a <italic>Mycobacterium marinum</italic>‐like mycobacterium was isolated from the yellowtail, <italic>Seriola quinqueradiata</italic>. The species was identified as <italic>M</italic>. <italic>marinum</italic> by a commercial mycobacterial DNA‐DNA hybridization kit. Nevertheless, PCR restriction analysis of the DNA of its RNA polymerase β‐subunit gene definitively showed that this <italic>Mycobacterium</italic> sp. was <italic>M</italic>. <italic>ulcerans</italic>. PCR analysis revealed the genotypic characteristics of <italic>M</italic>. <italic>ulcerans</italic> in the <italic>Mycobacterium</italic> sp., only the <italic>mup053</italic> gene sequence being absent, as has been found previously in other piscine mycobacteria such as <italic>M. marinum</italic> strains DL240490 and DL045 and <italic>M</italic>. <italic>pseudoshottsii</italic>. With one exception, this <italic>Mycobacterium</italic> sp. and <italic>M</italic>. <italic>pseudoshottsii</italic> had identical 16S rRNA gene sequences, which is also probably true of <italic>M. marinum</italic> strains DL240490 and DL045. Similarly, according to comparisons of the 16S rRNA gene, ITS region, and <italic>hsp65</italic> gene sequences, this <italic>Mycobacterium</italic> sp. is more closely related to <italic>M</italic>. <italic>pseudoshottsii</italic> than to <italic>M. ulcerans</italic> or<abstract abstract-type="main" xml:lang="en"> <title>Abstract</title> <sec id="mim514-sec-0001" sec-type="section"> <p>In Japan, a <italic>Mycobacterium marinum</italic>‐like mycobacterium was isolated from the yellowtail, <italic>Seriola quinqueradiata</italic>. The nucleotide sequence revealed insertion of IS<italic>2404</italic>, the sequence of which is 1366 bp long. The novel single nucleotide polymorphisms identified in this region distinguished this <italic>Mycobacterium</italic> sp. from <italic>M</italic>. <italic>marinum</italic> strain DL240490 and <italic>M</italic>. <italic>pseudoshottsii</italic>. The present findings raise the possibility that these species have a common ancestor. Further studies are required to improve our understanding of the relationship between their geographical origin and genetic diversity.</p> </sec> </abstract> … (more)
