- <abstract abstract-type="main" id="fml12188-abs-0001"> <title>Abstract</title> <p>Conjugative plasmid transfer systems have been well studied, but very little is known about the recipient factors that control horizontal transmission. A self‐transmissible IncP‐9 naphthalene catabolic plasmid, NAH7, carries the <italic>traF</italic> gene, whose product is considered to be a host‐range modifier of NAH7, because its <italic>traF</italic> deletion mutant (NAH7dF) is transmissible from <italic>Pseudomonas putida</italic> to <italic>P. putida</italic> and <italic>Escherichia coli</italic> and from <italic>E. coli</italic> to <italic>E. coli</italic>, but not from <italic>E. coli</italic> to <italic>P. putida</italic>. In this study, transposon mutagenesis of <italic>P. putida</italic> KT2440 was performed to isolate the mutants that could receive NAH7dF from <italic>E. coli</italic>. The mutants had the transposon insertions in <italic>ptsP</italic> or <italic>ptsO</italic>, encoding two of three components of the nitrogen‐related phosphotransferase system (PTS<sup>Ntr</sup>). The KT2440 derivative lacking <italic>ptsN</italic>, encoding the remaining component of PTS<sup>Ntr</sup>, was also able to receive NAH7dF. These results indicated that the PTS<sup>Ntr</sup> in <italic>P. putida</italic> is involved in inhibition of conjugative transfer of NAH7dF from <italic>E. coli</italic>.
