- <abstract abstract-type="main" id="pbr12046-abs-0001"> <title>Abstract</title> <p>Six novel high molecular weight glutenin subunits (HMW‐GS) from <italic>Aegilops speltoides</italic> (SS, 2n = 2x = 14) and <italic>Aegilops kotschyi</italic> (UUSS, 2n = 4x = 28) were identified by SDS‐PAGE and designated as <italic>ASy15*</italic>, <italic>AKx1*</italic>, <italic>AKx3*</italic>, <italic>AKx2.3</italic>, <italic>AKy20*</italic> and <italic>AKy8*</italic>, respectively. Their complete open reading frames (ORFs) were cloned and sequenced by allele‐specific PCR (AS‐PCR). Sequence comparison demonstrated that these novel genes displayed high single‐nucleotide polymorphisms (SNP) and InDel variations. In particular, <italic>AKy8*</italic> had an extra cysteine residue at position 140 in the central repetitive domain, while <italic>AKx2.3</italic> had an unusually long repetitive domain (816 aa), which might have positive effects on gluten quality. Phylogenetic analysis showed that <italic>AKx1*</italic> and <italic>AKx3*</italic> belonged to the 1S genome, <italic>AKx2.3</italic> to the 1U genome, and <italic>AKy20*</italic> and <italic>AKy8*</italic> most likely to the 1S genome. The divergence between the 6 HMW‐GS genes from the two <italic>Aegilops</italic> species and those from <italic>Triticum</italic> species occurred 4.77–23.54 MYA.
